Exercise cuts a gut metabolite that harms bones
Regular exercise is known to protect bones. But new research reveals a specific biochemical link: physical activity lowers a gut-derived metabolite called TMAO that fuels inflammation and accelerates bone breakdown.
TMAO (trimethylamine-N-oxide) is a compound produced by gut bacteria from dietary components such as choline and carnitine, found in meat and eggs. Higher TMAO levels in the blood have previously been linked to cardiovascular disease. What is new is that the researchers found a connection between regular physical activity, lower TMAO levels, reduced inflammation, and less harmful bone remodelling.
From gut microbiome to bone
The link runs through the gut microbiome: people who exercise regularly have a different composition of gut bacteria. That bacterial composition leads to lower TMAO production. Less TMAO means fewer inflammatory signals in the blood, with measurable effects on bone tissue. Bone remodelling processes, the normal balance between bone breakdown and bone formation, were less disrupted in people with lower TMAO levels.
This is an association study: it shows correlations, not a proven causal chain. Whether reducing TMAO through other means, such as diet or probiotics, would have the same effect on bone has not been established.
Three processes connected
What makes this research noteworthy is that it links three processes studied separately in the aging literature: physical activity, gut microbiome composition and bone health. All three decline with age. This study suggests they may be connected through a single biochemical link.
From a longevity perspective, this is relevant: exercise has effects well beyond muscle maintenance and cardiovascular fitness. It influences what happens in the gut and bloodstream, with consequences reaching the skeleton. Whether the type, intensity or timing of exercise matters for TMAO and bone specifically has not yet been investigated.
