ABT-263 speeds up wound healing in aging skin

Aging skin heals more slowly -- that much has long been known. But a new experiment shows that clearing out damaged cells can significantly speed up the process.

Researchers tested ABT-263, a compound that selectively eliminates cells that have stopped dividing -- known as senescent cells. These cells accumulate as you age and actively interfere with tissue repair. In elderly mice, wounds were treated with ABT-263 applied directly to the skin as a cream. The result: wounds healed considerably faster than in untreated animals.

The study also shows that ABT-263 switches on genes involved in collagen production and tissue repair. That suggests the compound does more than just clear out unwanted cells -- it may also actively kick-start the healing process.

Senescent cells as a brake on recovery

Senescent cells are not simply inert debris. They secrete substances that affect surrounding cells and keep inflammatory responses simmering. The more senescent cells are present, the less room there is for new, healthy cells to grow. In aging skin, that effect shows up clearly as slower wound healing.

ABT-263 belongs to a class of compounds called senolytics, which specifically target senescent cells. Earlier research tested these substances mainly through injection or oral administration. The new approach -- delivering the compound via a cream -- is notable because it works locally, limiting the risk of systemic side effects.

From mice to humans: still a big leap

The results are promising, but the research was carried out in mice. Translating those findings to humans is far from straightforward. Human skin differs meaningfully from mouse skin in thickness, cell composition, and the mechanisms it uses to heal. On top of that, ABT-263 was originally developed as a cancer drug and is known to cause side effects at higher doses.

Even so, the study points in an intriguing direction. If senolytics can be applied locally without triggering systemic effects, that opens the door to potential treatments for people who struggle with slow wound healing -- such as older adults or patients with diabetes.
