Better brain drainage cuts tau buildup in mice

Your brain has its own cleaning system that flushes out waste, mostly while you sleep. In older people, this system gradually fails, and that may contribute directly to the protein buildups seen in Alzheimer’s disease.

The system in question is the glymphatic network: a set of channels that drives cerebrospinal fluid through brain tissue, carrying away metabolic waste in the process. A key protein in this network is aquaporin-4 (AQP4), which acts as a water channel in brain cell membranes. As people age, glymphatic flow declines. Waste accumulates, promoting inflammation and cell dysfunction.

Researchers tested whether a small molecule called TGN-073 could boost AQP4 activity and restore drainage. The study used PS19 mice, a genetically modified model for tau protein accumulation, a hallmark of Alzheimer’s disease and related conditions. The animals were treated with TGN-073 over an extended period.

Less tau, less cell death

The results were encouraging. Treated mice showed measurable improvements in fluid movement through brain tissue. Tau levels dropped both in the brain itself and in cerebrospinal fluid. Neurodegeneration, assessed by markers of cell death, was also reduced compared to untreated animals. Exactly how TGN-073 increases AQP4 activity remains unclear. It may alter the protein’s structure, widening the pores it creates in cell membranes.

It is important to stress that this is animal research. Whether TGN-073 is safe or effective in humans is not yet known. The mice used are also a model of genetic tau pathology, not normal aging. That said, the findings suggest that glymphatic dysfunction may play a causal role in tau accumulation, rather than being merely a secondary feature of disease.

Why sleep matters for the aging brain

From a longevity perspective, this research is notable for what it implies more broadly. The glymphatic system is most active during sleep. Chronic sleep deprivation may structurally weaken brain drainage over time. If activating AQP4 confers a protective effect, it may also help explain why good sleep is consistently associated with lower dementia risk. The findings were reported via Fight Aging, drawing on work published in peer-reviewed literature.
