Five blood proteins flag liver disease sixteen years early
A liver disease linked to obesity and poor diet is usually caught only after serious damage has occurred. A panel of five blood proteins could change that timeline dramatically.
Researchers publishing in Nature Aging used plasma proteomics, the large-scale measurement of proteins circulating in the blood, to identify a five-protein panel that predicts future risk of metabolic liver disease. The study reports that this panel can flag risk up to sixteen years before clinical onset of MASLD (metabolic dysfunction-associated steatotic liver disease).
What is MASLD and why does early detection matter?
MASLD is a condition in which fat accumulates in the liver independently of alcohol consumption. It is closely tied to insulin resistance and obesity, and it progresses silently for years. By the time symptoms appear, significant liver damage may already be present.
The researchers analysed stored blood samples and identified which combination of proteins best predicted who would go on to develop MASLD years later. The five-protein panel showed sufficient predictive power to function as a risk stratification tool, a way of sorting people by their likelihood of developing disease before it becomes clinically apparent.
A sixteen-year window for prevention
A prediction horizon of sixteen years is unusually long. If these findings hold up in larger, independent cohorts, they could enable early lifestyle interventions before organ damage occurs. The researchers describe the panel as a potential tool for early risk stratification, though they emphasise that further validation is needed before clinical recommendations can be made.
For longevity research, this is meaningful: liver disease accelerates biological ageing and is associated with a wide range of metabolic conditions. Blood protein screening fits within a broader shift towards preventive medicine focused on catching disease long before symptoms emerge.
