Shingles vaccine linked to 24% lower dementia risk

A vaccine against shingles appears to do more than protect against a painful rash. In a study of over 500,000 older adults, those who received the Shingrix vaccine were 24% less likely to be diagnosed with dementia over four years.

Shingles is caused by the varicella-zoster virus, which lies dormant in nerve cells after a chickenpox infection and can reactivate later in life. The researchers found that vaccination against shingles was associated with a substantial reduction in dementia diagnoses. Roughly one dementia case was prevented for every 17 people vaccinated.

The mechanism behind this association is not yet established. One hypothesis is that the herpes virus, when reactivated, triggers chronic low-grade inflammation in nerve tissue. This type of persistent inflammation, known as inflammaging, has been linked to multiple age-related diseases, including neurodegenerative conditions. Suppressing viral reactivation might reduce that inflammatory burden on the brain over time.

A large sample, but observational caveats remain

The scale of the study is notable. Half a million participants is exceptional for this type of research. However, this was an observational study, meaning people who choose to get vaccinated may differ in other health-relevant ways from those who do not. The researchers corrected for potential confounding factors, but a causal link cannot be confirmed without a randomised controlled trial.

Vaccines as a longevity tool?

From a longevity perspective, the finding is worth attention. An existing, widely available and safe vaccine may offer benefits that extend beyond its primary indication. Earlier research has suggested a link between certain viral infections and elevated dementia risk. This study adds a concrete figure to that line of evidence, while the researchers themselves note that further verification is needed.

The result fits a growing body of work suggesting that chronic infection and inflammation play a larger role in brain ageing than previously assumed.
